Membrane-Bound Adenosine Deaminane from Human Placenta



Abstract
The properties and activity change with gestational age of plasma membrane bound adenosine deaminase from human placenta have been investigated.
1. The enzyme exhibited a broad pH optimum from pH 6 to 9 and is heat labile, being completely inactivated by heat treatment at 70 ¡ÆC for 10 minutes.
2. The enzyme is completely inhibited by 2 mMCu2+ and recovered by EDTA(4 mM) only in case of adding Cu2+ after preincubation of enzyme with substrate.
3. The apparent Michaelis constant of enzyme for adenosine is 31 mM and for deoxyadenosine is 26 mM.
4. The enzyme is not affected by concanavalin-A.
5. The specific ADase activity increased with gestational age by about 2-fold.
It is discussed that the enzyme is strongly related to cellular differentiation of placenta and is different from Abase of soluble fraction of human placenta in its biologic role.
